Btw, they have not signed the contract even though there is a place for them to sign on the contract. But, I do have numerous emails from the rental manager confirming the event.

Hello,
If I was given a contract by a rental agency to hold an event at their location, and I signed it and submitted payment for the rental, is it possible for them to cancel the contract?
They have not cashed my check, yet. But, they did accept my signed contract.
Thank you!

If you signed it and submitted payment for the rental than it is not legal for them to cancel the contract. Its totally unfair. You need to report them if they are not accepting your signed contract...
